Enzyme action and competitive inhibition
Raise the substrate concentration and see the reaction rate level off. Add a competitive inhibitor and compare.
What to notice
A competitive inhibitor resembles the substrate and competes for the active site; enough substrate can outcompete it.
Key relation
v = V_max[S] / (K_m + [S]) ; competitive: K_m(app) = K_m(1 + [I]/K_i)
Try this
Add inhibitor, then raise [S] a lot. Does V_max change?
What you can change
- Substrate concentration [S]: 0 to 50 mM
- Michaelis constant K_m: 1 to 20 mM
- Maximum rate V_max: 10 to 100 μmol/min
- Competitive inhibitor [I]: 0 to 20 mM
